Miles Morales is een leerling die bij zijn moeder Rio Morales woont, zij is een verpleegster en zijn vader, Jefferson Davis, is een politie agent.

Nadat Oscorp-wetenschapper Dr. Conrad Markus het bloed van Peter Parker heeft gebruikt om de formule die Spider-Man heeft gemaakt na te maken. Deze formule zit wederom in een spin. De Prowler (Aaron Davis) steelt de formule, en tijdens de diefstal kruipt een van de spinnen in de plunjezak van de Prowler.

Dagen later wordt de neef van de Prowler, Miles Morales, tijdens een bezoek aan Aarons appartement door de spin gebeten. Morales ontwikkelt bovenmenselijke vermogens die vergelijkbaar zijn met die van Peter Parker, maar vertelt het zijn ouders niet, vanwege het wantrouwen van zijn vader jegens superhelden, en vertrouwt alleen zijn beste vriend, Ganke Lee.

Miles, die gewoon een normaal leven wil, is ongelukkig met het hebben van deze vermogens, en aanvankelijk misselijk bij het idee zijn leven te riskeren voor superhelden. Echter, na getuige te zijn geweest van de dood van Spider-Man door de handen van de Green Goblin, beseft Miles dat hij had kunnen helpen. Hierdoor werd Miles geïnspireerd om toch de misdaad te gaan bestrijden.

Als het Venom symbioot bezit neemt van Miles Morales krijgt hij nog meer extreme krachten
